My favourite card-based game ever is Metal Gear Acid (Playstation Portable). I would probably take a lot of gameplay cues from that, using an overhead view (or isometric) grid-based playing field.
In the past I had a game concept in mind, and... it was squirrel-based. A big part of it would be collecting items, which in this case could be the cards. The conflict would be competition against other squirrels, kinda fighting over resources. It's pretty vague, but it's a starting point.
